Role of Estrogen Receptor Alpha rs3798577 Polymorphism in Breast Carcinoma Risk Determination

BACKGROUND: Interaction between estrogen and receptor (ER) takes part in the regulation differentiation of breast tumorigenesis. Some ERα polymorphisms, including rs3798577, are reported to be associated with risk aggressiveness carcinoma since site was targeted by microRNA, which can further modulate expression. Hence, this study conducted disclose possible role SNP rs3798577 on patients.METHODS: Samples were taken from post-mastectomy tissues female patients screened based completeness medical histopathological records. DNA isolation proceeded using real time-polymerase chain reaction (RT-PCR) then analyzed for high resolution melting (HRM). The nucleotide base sequence polymorphism. ER immunohistochemistry test carried out counted quantitatively staining intensity percentage stained cells.RESULTS: Out 65 samples, there 33 samples as wild type 32 variant type. Most had >80% percentage. middle intensity, while strong intensity. Higher (52.2%) found weak histoscore, meanwhile higher (52.4%) but not significant (p=0.725).CONCLUSION: a lower weaker histoscore compared type, suggesting that polymorphism might play determination.KEYWORDS: cancer, ERα, polymorphism, immunoexpression
